Weed out the fonts you don't really need. You can still keep them but when a program like Illustrator refuses to load it's because it takes tremendous memory to run all those fonts. The same problem occurs with the type tool in Painter. If you can live with about 80 fonts in total, the program should run.
You need to obtain a font management program Adobe Type Manager that lets you load and unload fonts at wil from any directory in your system while keeping your Windows fonts folder clean of anything that Windows itself doesnt need.
