Maybe you can try ftp? both explorer and firefox have something like this via ftp. If not I'm sure there are ad ons for browsers which let you do this, but this is the javascript forum, not the java forum, or add on forum. Anyway any add-on or applet will mean more trouble for the end user. What functionality are you trying to to get perhaps there is a better way than drag and drop.
Given you are asking in the JavaScript forum, the answer is "Using JavaScript, cross browser and/or cross OS, this cannot be done".
Using Java (which is entirely different from JavaScript) may provide a different answer, but you should take your question to the Java forum if you want to know about this.

P.S. You could always investigate (and ask in the appropriate forum for help with):
WebDAV (e.g. Jakarta Slide)
SWFUploader (for uploading images using a Flash movie)
I've investigated both of these quick a lot over the last 2 months or so, and we ended up going with the latter. The former was too much like hard work for us, although it was a hell of a lot more configurable and gave much more flexibility in terms of UI.
