Should I run a defragmenting tool like diskkeeper on my SQL data files? I am running a sql 2005 database and Windows 2003 SP2 OS.

No, it won't be able to do anything. SQL Server locks the files so that nothing else can access them.

If you need to defrag the databases you'll need to stop the SQL Service, then defrag the files.

This will give you a small performance benefit if anything. It usually isn't worth the downtime.
