Ok. Here I go again. How would I go about figuring out disk layout & filesystem layout if I were to start to make an email server? Which resources should I read?

Well, al depends on what you are going to use. If the Maildir option is used then /home may well need to be a reasonable size as the mail will be in the /home/<user> directories.
If mailboxes are to be used then /var will be impacted. (/var/spool/mail)
In general /var needs to be of a decent size, it doesn't matter if you are using sendmail, qmail or postfix, they all either live and spool in various directories in /var.

As an example we have allocated 10Gb to /var as we had lots of disk space to play with, plus we use other packages like MQSeries which uses /var as well.
