Difference between except and different from operators

I am creating a query where I select and object A
conditions are
Ist set
A="U" or A different from "U"
2nd set
A="U" or A except "U"

I am not getting any data for the 2nd set.
I feel the result should be same for both the queries.Can anybody throw some light on this?
