Denied Acces to Drives on Primary Domain Controller

Why do my drive mappings (U and S) deny my access into those drives which are placed on the primary domain controller? My users can log in successfully in the morning but as the day goes on, and they try to get back into one of the drives, they are denied acces and are forced to reboot in order to reconnect. The drives don't seem to be disconnected because no RED X is seen through the drives...but access is still denied. I have turned off auto disconnect on the server but still seem to have the problems. This primary domain controller is in addition to 2 other servers (secondary domain and backup) which I don't have a problem with.
