My boss has a Windows XP Pro home computer connecting to a Windows XP Pro computer in the office. Is it possible to set the default printer in the office to the network printer in the office and the default for the home PC to the printer attached to the home PC? Or does she have to keep selecting the proper printer each time?

How does he connect to the Windows XP Pro computer in the office?

She connects through remote access via the internet.

Maybe try creating another login on the work PC, and that would allow you to set two different default printers. Of course, the new user account would need acces to all the same resources, but if they're network file shares that's no problem.
