Create a New Data Store or Storage Group?

I am out of disk space on our Exchange server. I added a new drive in and want to move some users to the new drive.Should I go about creating a new data store or storage group?
Also, is it possible to have one storage group or data store online while defragging the other? My plan is to create either a new storage group or data store on the new drive,move some users to the new storage group, and while the new storage group is online ,defrag the other so that some users could have email access

Other question is do you have Exchange Enterprise Edition or Standard?

Only Enterprise lets you create multiple storage groups.
