Use either 'interactive' transparency tool or Effects-Lens-transparency (choose from drop down list). Be aware that Pantone spot colours will be changed to cmyk, this is obviously an issue if separating for output. If you find that they are not separating corectly in cmyk try printing without using 'print preview'
