Connecting Exchange 2000 to Exchange 5.5

I am working with another company to get an SMTP connector set up between our 2 organizations. I have Exchange 2000, so I will be using an SMTP connector on my end to make this happen. I have done this with another domain, so I know my configuration is correct; however, the other company has Exchange 5.5. I am trying to help him out by configuring his mail server for him, but I cannot recall how to route email for a specific domain to a particular IP address. Do you add my domain to the "Specify by E-mail domain" button on the Connections tab of the IMC? This seems most logical, but it does not work. Any help would be greatly appreciated.
