connect to upstream proxy

I have a computer room that i am configuring! We have installed isa server as the local proxy server, all the boxes are able to access the internet via an automatic configuration script, that is pointing to an upstream proxy on the other end of the link! How do i set up the local proxy to access the upstream proxy? How do i setup each pc to access the local proxy? At present the local proxy is turned off!
Any help would be greatly appreciated!
