I have a black and red spot color document in Quark. I want to convert it to a PDF file to send out to printing place. Will the PDF file separate into two-colors properly? Thank you.

Only if you use the "DeviceN" option instead of "Combined CMYK" in the output tab of the Quark print window.
And use the distiler making sure you are compatible with at least Acrobat V6.

And only if your printing place is compatible with Postscript Level 4

Otherwise print to PDF using Seperations from Quark.
