How could I change a greyscale picture to use a pantone colour in Indesign or Ilustrator???

In Illustrator, use opacity masks.

1: Draw a shape large enough to cover your image, and fill it with your pantone colour.
2: Bring the image in front of this shape.
3: Select both items, then go to the Transparency palette.
4: From its menu, select "Make Opacity Mask" (make sure "Invert Mask" is selected).
