One of our software applications is experiences slowness after this passed weekend. I have been told that no changes have been made to any server. Is there actually a way to prove this (changes made or not) other than I guess checking the modified date of any files?

Start off with checking with the application and system logs for the weekend to see if they provide any information (like any antivirus updates, patches, etc).
Take a look at the task manager to see if you have any memory leaks, page faults, processes that are taking up the CPU time etc.
Have you also tried performing a reboot to see if it clears up the issue?

Yes, we have rebooted multiple times. I have scanned through the event viewer but can't see anything.
