Programs will be just as effective running with others and by themselves. However running multiple programs at once will cause them to take more time to scan because there all fighting over processor/ram . Plus one program might come across what another one has found also. Anyways yeah they can be ran together, I have a friend who does that all the time.
I do it, too. I have heard, however, that some AV programs don't like it. I've used Sophos and e-Trust (AV programs) with Ad-aware, MS Antispyware, and Spy-bot (AS programs) with no problems. I just don't run all 5 at the same time. ;-)

With programs that run in the background and perform active protective functions, you can run into conflicts. Generally, you do not want to run two AV or two AS programs at the same time. One of each type however, should work fine since they are looking for different things. I run AVG and MS Anti-Spyware together with no problems.
