Altering the layout using CSS

Hi,
I can't seem to get my head around this. I know that I can create a CSS file with definitions and layout features for DIVs with a .CSS extension and then link it to a page.
What I cant understand is the claim that the layout can be changed by altering the CSS file. Sure the layout will change, but doesnt it ultimately depend of the html file and how its structured. ie: how the Divs are placed in the HTML code?
Please, if any one can clarify this.. its much appreciated.
