I will be setting up a new location with MM (5.2) when their office moves. They will be getting new phone numbers. If I set up the MM boxes now and put their new extension in the MM AD tab, later when the office moves and all of their phone numbers change, will that affect the MM tab at all on the extension field? Or will the info I have in there remain as is?

Active Directory-Phone Number Change

AD phone number is only used in an attempt to prepopulate the MM tab when enabling the account for the first time. Changes to those AD properties don't effect the MM subscriber (or what number they are assigned to.
Assuming their extension lengths are staying the same, I would run the vmenable tool to just change their extension/mailbox/numeric addresses once moved. Don't forget to change other extension related information in the VMD (operator extension, etc).
