Try using just HTTP versus HTTPS in the URL.
If no luck there, remember that the install of SUS includes the install of the IIS lockdown tool and you will have to add your own SUS site in to your trusted sites within Internet Explorer to access the admin pages and then the HTTPS should work but...

There's a lot of things that can result in this error. Couple o' questions:
Is the E2K server a DC? Do you have any other W2K servers on the domain with AD running? Do you have the [P]DC as the GC and master for the domain?
E2K isn't recommended for installation on a DC. It works...
