Depending on what software you are using there is a special Echange Backup Agent which will do a full backup of exchange and get the file normal backup misses. This is important as I found out the hard way. We use ArcServe IT but Veritas also has an Exchange Backup Agent.
Client Access Supports Native TCP/IP. I would advise against the 32 bit DLC (APPC) Connection.
On Client Access Connections just use the Connections Wizard (New Connection) with the IP Address of your AS/400 and it should work.
