I've set the DataFormatString on a gridview but nothing is happening

You need to add the following line above the DataFormatString

<asp:BoundField DataField ="TestMePercent">
HeaderText="TestMe" 
HtmlEncode="False"
DataFormatString="{0:p0}" />

HtmlEncode is a property that was added to prevent CSS attacks. Setting this property to false, allows the DataFormatString to be applied.
